Sekulow was born and brought up in Brooklyn, New York, and raised in a Jew family. However, he decided to convert to Christianity while in college and is currently a Messianic Jew.
Sekulow attended the Lakeside High School in Atlanta, Georgia. Immediately after completing high school, Jay joined Mercer University where he studied and earned a B.A. and a J.D. in 1977.
Sekulow got interested in Christianity and converted after encountering Jews for Jesus while attending Atlanta Baptist College now known as the Atlanta campus of Mercer University.
He earned a Ph.D. from Regent University in 2005, writing his dissertation on religious influence on Supreme Court Justices and their opinions.

In 1982, he started his own law company in Atlanta, Georgia. In 1987, he got inspired by the Jews and became a general counselor.
He established the not-for-profit group Christian Advocates Serving Evangelism in 1988.

In 1992, he became the director of ACLJ, where he worked as the main chief counselor and principal officer.
